Zileuton (Zyflo)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Zileuton

What it is

Zileuton is a 5-lipoxygenase inhibitor: it blocks the enzyme that makes leukotrienes, inflammatory signals that narrow the airways. It is used for long-term control of asthma in the US, as tablets taken several times a day or extended-release tablets. Liver tests are checked because it can raise liver enzymes. It is sold as Zyflo.

How genes come into it

Zileuton is broken down by CYP1A2, CYP2C9 and CYP3A4 and by glucuronidation. The gene study listed, a genome-wide study of asthma response to leukotriene medicines, looked at PRORP (MRPP3). The evidence is weak, and there is no CPIC or DPWG guideline.

What research found

Every published finding on gene variants and Zileuton. Upload your DNA file to see which of these rows apply to you.

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

PRORP rs12436663: Level 3. Efficacy: Asthma. 1.

Response (1 finding)

Studies of how well it worked in people carrying a variant.

PRORP rs12436663: Genotype AA is associated with decreased response to zileuton in people with Asthma as compared to genotypes AG + GG.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
