Saxagliptin is a DPP-4 inhibitor: it blocks the enzyme that breaks down incretins, gut hormones such as GLP-1 that make the pancreas release insulin after a meal. It is used for type 2 diabetes, alone or combined with metformin or dapagliflozin. It is sold as Onglyza.
Saxagliptin is broken down by CYP3A4 and CYP3A5 into an active product, so strong CYP3A4 blockers raise its levels. The gene studies listed looked at GLP1R and PRKD1 for how much blood sugar fell. The findings come from a few studies, and there is no CPIC or DPWG guideline.
