Risedronate (Actonel)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Risedronate

What it is

Risedronate is a bisphosphonate: it binds to bone and slows the osteoclasts that break bone down, so bone density is preserved and fractures become less likely. It is taken as a daily, weekly or monthly tablet for osteoporosis after the menopause, in men, and with long-term steroid use, and for Paget's disease of bone. It is sold as Actonel and Atelvia, among other names.

How genes come into it

Risedronate is not broken down; what bone does not take up is cleared by the kidneys. The few gene studies listing it looked at IL1B and at LRP5, a gene important for bone formation, for how much bone density improved. The findings come from a few studies, and there is no CPIC or DPWG guideline.

What research found

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

IL1B rs16944: Level 3. Efficacy: Osteitis Deformans. 1.

Other findings (1 finding)

Studies that do not fall into the groups above.

LRP5 rs3736228: Allele T is not associated with response to risedronate in people with Osteoporosis as compared to allele C.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
