Clodronate (Bonefos)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Clodronate

What it is

Clodronate is a first-generation bisphosphonate without a nitrogen group: it binds to bone and is taken up by osteoclasts, which it disables. It is used in Europe and elsewhere for bone damage and high calcium caused by cancer, especially multiple myeloma and breast cancer that has spread to bone; it is not approved in the US. It is sold as Bonefos and Loron.

How genes come into it

Clodronate is not broken down; what bone does not take up is cleared unchanged by the kidneys. The few gene studies listing it looked at VDR, the vitamin D receptor, and IL1B for bone response. The findings come from a few studies, and there is no CPIC or DPWG guideline.

Rated evidence (3 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

VDR rs731236: Level 3. Efficacy: Osteitis Deformans. 1.

IL1B rs16944: Level 3. Efficacy: Osteitis Deformans. 1.

VDR rs1544410: Level 3. Efficacy. 1.
