Dolasetron is an anti-sickness medicine of the 5-HT3 antagonist group: it blocks serotonin receptors in the gut and the brain's vomiting centre. It is used as tablets to prevent nausea and vomiting with chemotherapy; the injection is no longer used for this because of effects on the heart rhythm. It is sold as Anzemet.
Dolasetron is rapidly converted to hydrodolasetron, its active form, which is broken down partly by CYP2D6. A study looked at CYP2D6 for how well it prevented vomiting, and another at NOS1AP for effects on the QT interval of the heartbeat. CPIC reviewed dolasetron in its 5-HT3 antagonist guideline but made a recommendation only for ondansetron and tropisetron.
