Dimethyl Fumarate (Tecfidera)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Dimethyl Fumarate

What it is

Dimethyl fumarate is an oral immunomodulator, turned in the body into monomethyl fumarate, which activates the Nrf2 antioxidant pathway and shifts immune cells towards a calmer state. It is used for relapsing multiple sclerosis (Tecfidera) and, in Europe, for psoriasis (Skilarence). Flushing and stomach upset are common at first, and white cell counts are monitored.

How genes come into it

Dimethyl fumarate is broken down by esterases, not CYP enzymes. The gene study listed looked at GSTP1, a glutathione transferase, for response in multiple sclerosis. The evidence is weak, and there is no CPIC or DPWG guideline.

What research found

Every published finding on gene variants and Dimethyl Fumarate. Upload your DNA file to see which of these rows apply to you.

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

GSTP1 rs1695: Level 3. Efficacy: Psoriasis. 1.

Response (1 finding)

Studies of how well it worked in people carrying a variant.

GSTP1 rs1695: Genotype GG is associated with decreased response to dimethyl fumarate in people with Psoriasis as compared to genotypes AA + AG.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
