About Cavosonstat

What it is

Cavosonstat (N91115) was an experimental medicine for cystic fibrosis that inhibits S-nitrosoglutathione reductase, an enzyme thought to help break down the faulty CFTR protein. It was tested alongside ivacaftor and lumacaftor in people with the F508del variant, but did not show enough benefit and was not developed further.

How genes come into it

Cavosonstat was meant for cystic fibrosis caused by the F508del variant in CFTR, the gene behind the condition. The gene studies on it come from its clinical trials in people with F508del. There is no CPIC or DPWG guideline.

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

CFTR rs113993960: Level 3. Efficacy: Cystic Fibrosis. 1.

Response (2 findings)

Studies of how well it worked in people carrying a variant.

CFTR rs113993960: Genotype del/del is associated with response to cavosonstat in people with Cystic Fibrosis as compared to genotypes CTT/CTT + CTT/del.

CFTR rs113993960: Genotype del/del is not associated with response to cavosonstat in people with Cystic Fibrosis as compared to genotypes CTT/CTT + CTT/del.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
