Carboplatin (Paraplatin)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Carboplatin

What it is

Carboplatin is a platinum chemotherapy medicine, a gentler relative of cisplatin: it cross-links the strands of DNA so cancer cells cannot copy it. It is used for ovarian and lung cancers and many others, often combined with paclitaxel. It causes less kidney damage and hearing loss than cisplatin, but lowers blood counts more. It is sold as Paraplatin, among other names.

How genes come into it

Carboplatin is cleared unchanged by the kidneys, so its level depends mostly on kidney function rather than on genes. Studies have looked at DNA repair genes (such as XRCC1), growth-signal genes (AKT1, ERBB3, VEGFA) and transporters for response and side effects. The findings come from single or a few studies, and there is no CPIC or DPWG guideline.

Rated evidence (65 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

ERCC1 rs11615: Level 3. Toxicity: Neoplasms. 6.

MTHFR rs1801131: Level 3. Efficacy: Neoplasms. 4.

MTR rs1805087: Level 3. Efficacy, Toxicity: Neoplasms. 3.

SLCO1B3 rs4149117: Level 3. Toxicity: Lung Neoplasms. 2.

Response (26 findings)

Studies of how well it worked in people carrying a variant.

AKT1 rs2494752: Genotype AG is associated with decreased response to carboplatin or cisplatin in people with Carcinoma, Non-Small-Cell Lung as compared to genotypes AA + GG.

BARD1 rs2070096: Allele A is associated with decreased response to carboplatin, docetaxel and trastuzumab in women with Breast Neoplasms.

EIF3A rs3740556: Genotypes AA + AG are associated with increased response to carboplatin and cisplatin in people with Carcinoma, Non-Small-Cell Lung as compared to genotype GG.

EIF3A rs3740556: Genotypes AA + AG are associated with increased response to carboplatin and cisplatin in people with Lung Neoplasms as compared to genotype GG.

Side effects (1 finding)

Studies of side effects in people carrying a variant.

GSTP1 rs1695: Allele G is associated with decreased response to carboplatin and paclitaxel in people with Ovarian Neoplasms as compared to allele A.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
