Vincristine (Oncovin)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Vincristine

What it is

Vincristine is a vinca alkaloid chemotherapy medicine from the Madagascar periwinkle. It blocks the assembly of microtubules, so dividing cells cannot separate their chromosomes. It is a key part of treatment for acute lymphoblastic leukaemia, lymphomas (in CHOP) and many childhood cancers. Nerve damage (neuropathy) is its main dose-limiting side effect. It is sold as Oncovin, among other names.

How genes come into it

Vincristine is broken down mainly by CYP3A5 and CYP3A4 and pumped out of cells by ABCB1. Children of African ancestry, who more often carry working CYP3A5 copies, clear it faster and in studies had less nerve damage. A genome-wide study in children with leukaemia linked a CEP72 variant to more neuropathy. The findings come from a few studies, and there is no CPIC or DPWG guideline.
