Prednisolone

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Prednisolone

What it is

Prednisolone is a corticosteroid, the active form of prednisone, that calms inflammation and the immune system through the glucocorticoid receptor. It is used for asthma and COPD flare-ups, allergic reactions, rheumatoid arthritis, inflammatory bowel disease, autoimmune conditions and some blood cancers, and as eye drops. It is sold as Orapred, Prelone and many other names.

How genes come into it

Prednisolone is broken down by CYP3A4 and pumped out of cells by ABCB1. Studies have looked at ABCB1 variants for response and side effects in conditions such as nephrotic syndrome and inflammatory bowel disease, and at immune genes such as TLR4 and GATA3. The findings come from single or a few studies, and there is no CPIC or DPWG guideline.

Rated evidence (4 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

PNPLA3 rs738409: Level 3. Toxicity: Acute lymphoblastic leukemia. 2.

DSE;TSPYL1 rs3828743: Level 3. Efficacy: Prostatic Neoplasms. 1.

ABCB1 rs2229109: Level 3. Efficacy: Acute lymphoblastic leukemia. 1.

ABCB1 rs1045642: Level 3. Toxicity: Kidney Transplantation. 1.

Response (9 findings)

Studies of how well it worked in people carrying a variant.

ABCB1 rs1045642: Genotype AA is associated with decreased resistance to doxorubicin, methotrexate, prednisolone and vincristine in children with Precursor Cell Lymphoblastic Leukemia-Lymphoma as compared to genotype GG.

ABCB1 rs2229109: Genotype CT is associated with increased resistance to doxorubicin, methotrexate, prednisolone and vincristine in children with Precursor Cell Lymphoblastic Leukemia-Lymphoma as compared to genotype CC.

CD40 rs1883832: Genotype CC is associated with decreased clinical benefit to prednisolone in children with Thrombocytopenia as compared to genotypes CT + TT.

GATA3 rs3824662: Genotype AA is associated with decreased clinical benefit to prednisolone in children with Precursor Cell Lymphoblastic Leukemia-Lymphoma as compared to genotypes AC + CC.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
