Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.
Pramipexole is a dopamine agonist: it activates dopamine D2 and D3 receptors directly, standing in for the dopamine the brain lacks in Parkinson's disease. It is used for Parkinson's disease and for restless legs syndrome. Impulse control problems, such as compulsive gambling or shopping, are a known side effect. It is sold as Mirapex and Sifrol.
Pramipexole is not broken down by liver enzymes; the kidneys clear it unchanged. The single gene study listed looked at DRD3, the gene for one of the receptors it acts on, for how well it eased Parkinson's symptoms. The evidence is weak, and there is no CPIC or DPWG guideline.

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.
DRD3 rs6280: Level 3. Efficacy: Parkinson Disease. 1.
Studies of how well it worked in people carrying a variant.
DRD3 rs6280: Allele T is associated with increased response to pramipexole.
Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
