Pegaptanib (Macugen)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Pegaptanib

What it is

Pegaptanib was the first anti-VEGF treatment for the eye: an aptamer, a short strand of RNA that binds VEGF-165 and stops it from driving abnormal blood vessel growth. Injected into the eye, it was used for wet age-related macular degeneration, and has been replaced by more effective antibodies such as ranibizumab and aflibercept. It was sold as Macugen.

How genes come into it

Pegaptanib acts in the eye and is broken down by nucleases. The gene study listed looked at VEGFA, the gene for its target, for how much vision changed after treatment. The evidence is weak, and there is no CPIC or DPWG guideline.

What research found

Every published finding on gene variants and Pegaptanib. Upload your DNA file to see which of these rows apply to you.

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

VEGFA rs2010963: Level 3. Efficacy: Choroidal Neovascularization. 1.

Response (1 finding)

Studies of how well it worked in people carrying a variant.

VEGFA rs2010963: Genotype CC is associated with decreased response to bevacizumab, pegaptanib or ranibizumab in people with Choroidal Neovascularization as compared to genotypes CG + GG.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
