Lamivudine (Epivir)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Lamivudine

What it is

Lamivudine (3TC) is a nucleoside reverse transcriptase inhibitor: it is built into viral DNA as it is copied and stops the chain from growing. It is one of the most used HIV medicines, part of many combination tablets such as Triumeq and Dovato, and at a lower dose treats chronic hepatitis B. It is sold as Epivir, among other names.

How genes come into it

Lamivudine is cleared mostly unchanged by the kidneys, with the help of the transporters SLC22A2 (OCT2) and MATEs. Gene studies listing it were mostly done in people on combination HIV treatment and looked at ABCB1, ABCC1, CYP2B6 and HLA-B, findings that largely concern the other medicines. There is no CPIC or DPWG guideline.

What research found

Every published finding on gene variants and Lamivudine. Upload your DNA file to see which of these rows apply to you.

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

ABCC1 rs212091: Level 3. Efficacy: HIV infectious disease. 1.

Metabolism (1 finding)

Studies of how quickly the body processes this medication.

SLC22A2 rs145450955: Genotype GG is not associated with clearance of lamivudine in healthy individuals as compared to genotype AG.

Response (10 findings)

Studies of how well it worked in people carrying a variant.

ABCC1 rs212091: Genotypes CC + CT are associated with increased resistance to lamivudine, lopinavir, ritonavir and zidovudine in people with HIV Infections as compared to genotype TT.

CYP2B6 rs2279343: Genotypes AG + GG is associated with increased clinical benefit to efavirenz, lamivudine and tenofovir in people with HIV Infections as compared to genotype AA.

HLA-B HLA-B*40:01: HLA-B *40:01 is associated with response to lamivudine, nevirapine and stavudine in people with HIV Infections.

ABCB1 rs1045642: Allele A is associated with response to lamivudine or nevirapine in people with HIV Infections as compared to allele G.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
