Cerivastatin was a highly potent statin that blocked HMG-CoA reductase, the liver enzyme that makes cholesterol. It was withdrawn worldwide in 2001 after reports of fatal rhabdomyolysis (severe muscle breakdown), especially when combined with gemfibrozil. It was sold as Baycol and Lipobay.
Studies of people who had severe muscle damage on cerivastatin looked at SLCO1B1, the transporter that carries statins into the liver, whose variants are now known to raise statin levels and muscle risk with simvastatin and others, and at RYR2. Cerivastatin was broken down by CYP2C8 and CYP3A4. As the medicine is no longer available, there is no CPIC or DPWG guideline.
