Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.
Busulfan is an alkylating chemotherapy medicine that cross-links DNA. At high doses by drip, it is used to wipe out the bone marrow before a stem cell (bone marrow) transplant; as tablets, it was long used for chronic myeloid leukaemia. Its blood levels are often measured during conditioning, because exposure strongly affects both side effects and success. It is sold as Busulfex, Myleran and Busilvex.
Busulfan is inactivated mainly by joining it to glutathione, done largely by the enzyme GSTA1. Studies have linked GSTA1 variants, and to a lesser extent GSTM1, to how fast busulfan is cleared, and CTH to side effects. The findings come from a few studies, mostly in children before transplant, and there is no CPIC or DPWG guideline.
