About Caffeine Jitters

Caffeine works by blocking adenosine receptors, the brakes that build up drowsiness across a waking day. ADORA2A codes for one of those receptors. This variant does not change the receptor protein itself but sits within the gene and travels with versions that appear to differ in how strongly they respond. It separates the question of how fast you break caffeine down from the separate question of how much you feel it, which is why some people clear caffeine quickly yet still feel wired.

Good to know: How wired caffeine leaves you is separate from how fast you clear it. The studies behind this variant are small and inconsistently replicated. Habitual intake and sleep debt shift caffeine response more than this position does.

Fun fact: Caffeine does not add energy at all; it blocks the receptor that reports how long you have been awake.

Discovery: Variation in this receptor gene was linked to differences in caffeine response in studies from 2003 onward.

The Variant

The single variant behind this trait

Gene: ADORA2A

Variant (rsID): rs5751876

Location: Chr 22:24,441,333

dbSNP rs5751876

The studies

A genetic variation in the adenosine A2A receptor gene (ADORA2A) contributes to individual sensitivity to caffeine effects on sleep (Rétey JV et al., Clinical pharmacology and therapeutics, 2007)

Effect of ADORA2A Gene Polymorphism and Acute Caffeine Supplementation on Hormonal Response to Resistance Exercise: A Double-Blind, Crossover, Placebo-Controlled Study (Rahimi MR et al., Nutrients, 2024)

Sleep Quality and Duration in Children That Consume Caffeine: Impact of Dose and Genetic Variation in ADORA2A and CYP1A (Jessel CD et al., Genes, 2023)

A genetic variation in the adenosine A2A receptor gene contributes to variability in oscillatory alpha power in wake and sleep EEG and A1 adenosine receptor availability in the human brain (Tichelman NL et al., NeuroImage, 2023)

Other lifestyle traits

Odor After Certain Foods: A variant in a liver enzyme that clears a fishy-smelling compound produced when gut bacteria digest choline-rich foods like eggs, fish and beans.

Lactose Tolerance: Variants near the LCT gene that set whether you keep producing lactase, the enzyme for digesting milk, into adulthood.

Galactose Handling: A common change in GALT, the enzyme that processes galactose, half the sugar in milk. It marks a lower-activity version without settling whether you carry it.
