Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Ventolin

What it is

Salbutamol (albuterol) is a short-acting beta-2 agonist: it activates beta-2 receptors on the smooth muscle of the airways, relaxing them within minutes. Inhaled, it is the standard reliever for asthma attacks and COPD symptoms and is used before exercise to prevent wheezing. It is sold as Ventolin and ProAir, among other names.

How genes come into it

Salbutamol is cleared mainly by sulphate conjugation (SULT1A3) and the kidneys. The most studied gene is ADRB2, the beta-2 receptor it acts on: its Arg16Gly variant was linked in some studies to a different response with regular use, but later trials did not confirm a clinically useful effect. Genome-wide studies flagged CRHR2, SPATS2L and others for bronchodilator response, more often in children of African and Latino ancestry. There is no CPIC or DPWG guideline.

Rated evidence (21 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

ADRB2 rs1042713: Level 3. Efficacy: Asthma. 5.

CRHR2 rs2267715: Level 3. Efficacy: Asthma. 1.

rs9552679: Level 3. Efficacy: Asthma. 1.

rs1423515: Level 3. Efficacy: Asthma. 1.

Response (39 findings)

Studies of how well it worked in people carrying a variant.

rs116551936: Genotype AG is associated with increased response to salbutamol in children with Asthma as compared to genotype GG.

rs17834628: Allele A is associated with increased response to salbutamol in children with as compared to allele G.

rs35661809: Allele G is associated with increased response to salbutamol in children with as compared to allele A.

rs7081864: Allele A is associated with decreased response to salbutamol in children with Asthma as compared to allele G.

Other findings (3 findings)

Studies that do not fall into the groups above.

ADRB2 rs1042713: Genotype AA is associated with decreased response to salbutamol in healthy individuals as compared to genotypes AG + GG.

COL2A1 rs3809324: Genotypes GT + TT is associated with decreased response to salbutamol in children with Asthma as compared to genotype GG.

VEGFA rs3025039: Genotypes CT + TT is associated with decreased response to salbutamol in children with Asthma as compared to genotype CC.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
