Rifapentine (Priftin)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Rifapentine

What it is

Rifapentine is a long-acting relative of rifampicin: it blocks bacterial RNA polymerase. Taken once a week together with isoniazid for twelve weeks (the 3HP regimen), or daily for one month, it treats latent tuberculosis infection, and it is used in some regimens for active tuberculosis. It is sold as Priftin.

How genes come into it

Rifapentine is cleared mainly by esterases and excreted in bile. The few gene studies listing it looked at ABCB1 for its levels and at NAT2, which concerns the isoniazid it is combined with. The findings come from a few studies, and there is no CPIC or DPWG guideline.

Metabolism (2 findings)

Studies of how quickly the body processes this medication.

ABCB1 rs1045642: Genotypes AA + AG is associated with increased clearance of rifapentine as compared to genotype GG.

NAT2 NAT2 slow acetylator: NAT2 slow acetylator is associated with increased concentrations of rifapentine in people with HIV Infections.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
