Pazopanib is a targeted cancer medicine, a tyrosine kinase inhibitor: it blocks several growth-signal receptors, chiefly the VEGF receptors that tumours use to grow new blood vessels. It is used for advanced kidney cancer and for some soft tissue sarcomas. It is sold as Votrient.
The US FDA lists two inherited genes for pazopanib, both about side effects on the liver. People who carry HLA-B*57:01 had a higher rate of raised liver enzymes in clinical trials, and people with the low-activity UGT1A1 version behind Gilbert's syndrome (two copies of *28) more often had raised bilirubin, because pazopanib also blocks the enzyme UGT1A1. There is no CPIC or DPWG guideline. Studies have also looked at the VEGF pathway genes VEGFA and KDR, with weaker evidence.
Clinical guidelines that name a gene for Pazopanib. They sort people into groups by the gene copies they carry.
US FDA: HLA-B. *57:01 positive. Data that Indicate a Potential Impact on Safety or Response.
US FDA: UGT1A1. Poor Metabolizer. Data that Indicate a Potential Impact on Safety or Response.

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.
UGT1A1 UGT1A1*1, UGT1A1*6, UGT1A1*28, UGT1A1*37: Level 3. Toxicity: Renal Cell Carcinoma. 2.
KDR rs34231037: Level 3. Efficacy, Metabolism/PK: Kidney Neoplasms. 1.
