Panitumumab (Vectibix)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Panitumumab

What it is

Panitumumab is a biologic medicine, a fully human monoclonal antibody that blocks EGFR, a growth-signal receptor on many cancer cells. Given by drip every two weeks, it is used for advanced bowel cancer, alone or with chemotherapy. A skin rash is its typical side effect. It is sold as Vectibix.

How genes come into it

Whether panitumumab works in bowel cancer depends on the tumour: it does not work when the cancer carries mutations in KRAS or NRAS, so tumour tissue is tested before treatment. Those mutations are acquired in the cancer, not inherited. For inherited genes, studies have looked at the EGFR ligands AREG, EREG and EGF and at the antibody receptor FCGR2A, with mixed results. There is no CPIC or DPWG guideline.

Rated evidence (6 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

KRAS rs61764370: Level 3. Efficacy: Neoplasms. 6.

AREG rs1353295: Level 3. Efficacy: Colorectal Neoplasms. 1.

AREG rs13104811: Level 3. Efficacy: Colorectal Neoplasms. 1.

EGFR rs712829: Level 3. Efficacy: Colorectal Neoplasms. 1.

Response (32 findings)

Studies of how well it worked in people carrying a variant.

AREG rs11942466: Genotype AA is associated with decreased response to cetuximab or panitumumab in people with Colorectal Neoplasms as compared to genotypes AC + CC.

AREG rs13104811: Genotype GG is associated with increased response to cetuximab or panitumumab in people with Colorectal Neoplasms as compared to genotypes AA + AG.

AREG rs1353295: Genotype GG is associated with increased response to cetuximab or panitumumab in people with Colorectal Neoplasms as compared to genotypes AA + AG.

AREG rs9996584: Genotype AA is associated with increased response to cetuximab or panitumumab in people with Colorectal Neoplasms as compared to genotypes AG + GG.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
