Osimertinib (Tagrisso)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Osimertinib

What it is

Osimertinib is a third-generation EGFR tyrosine kinase inhibitor designed to block both the common activating EGFR mutations and T790M, the mutation that makes earlier EGFR inhibitors stop working. It is used for EGFR-mutated non-small cell lung cancer, as first treatment, after surgery and after earlier EGFR inhibitors, and it reaches the brain well. It is sold as Tagrisso.

How genes come into it

Whether osimertinib works depends on EGFR mutations in the tumour (exon 19 deletions, L858R, T790M), found by testing tumour tissue or tumour DNA in the blood; these are acquired in the cancer, not inherited. The single gene study listed looked at LZTR1 in tumours. Osimertinib is broken down mainly by CYP3A4. There is no CPIC or DPWG guideline.

Response (1 finding)

Studies of how well it worked in people carrying a variant.

LZTR1 rs1411200130: Allele G is associated with increased resistance to osimertinib in women with Non-Small Cell Lung Carcinoma.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
