About NNRTIs

What it is

Non-nucleoside reverse transcriptase inhibitors (NNRTIs) are a class of HIV medicines that bind reverse transcriptase near its active site and block it, so HIV cannot copy its genetic material into the cell's DNA. The class includes efavirenz, nevirapine, rilpivirine, etravirine and doravirine, always used with other HIV medicines. This page collects studies on NNRTIs as a group.

How genes come into it

The key gene for the older NNRTIs is CYP2B6, which breaks down efavirenz and nevirapine: people with slow CYP2B6 versions, such as *6, reach much higher levels and more side effects. CPIC and DPWG have published guidelines on CYP2B6 and efavirenz, on its own page. The gene studies listed for the class concern CYP2B6.

Response (5 findings)

Studies of how well it worked in people carrying a variant.

CYP2B6 rs3745274: Allele T is associated with increased response to efavirenz or non-nucleoside reverse transcriptase inhibitors in women with HIV Infections as compared to genotype GG.

CYP2B6 rs1042389: Allele C is not associated with response to efavirenz or non-nucleoside reverse transcriptase inhibitors in women with HIV Infections as compared to genotype TT.

CYP2B6 rs28399499: Allele C is not associated with response to efavirenz or non-nucleoside reverse transcriptase inhibitors in women with HIV Infections as compared to genotype TT.

CYP2B6 rs34097093: Allele C is not associated with response to efavirenz or non-nucleoside reverse transcriptase inhibitors in women with HIV Infections as compared to genotype TT.
