Nitroprusside (Nipride)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Nitroprusside

What it is

Sodium nitroprusside is a powerful vasodilator that releases nitric oxide, relaxing both arteries and veins within seconds. Given by continuous drip in intensive care, it lowers blood pressure rapidly in hypertensive emergencies and during some surgery, and helps in acute heart failure. Its breakdown releases cyanide, so the duration and amount are limited. It is sold as Nipride and Nitropress.

How genes come into it

Nitroprusside breaks down in the blood, releasing nitric oxide and cyanide, which the liver converts to thiocyanate. The gene study listed looked at ACE for blood vessel responses to nitroprusside in research settings. The evidence is weak, and there is no CPIC or DPWG guideline.

What research found

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

ACE rs1799752: Level 3. Efficacy. 1.

Response (1 finding)

Studies of how well it worked in people carrying a variant.

ACE rs1799752: Allele ATACAGTCACTTTTTTTTTTTTTTTGAGACGGAGTCTCGCTCTGTCGCCC is associated with increased response to acetylcholine or nitroprusside in healthy individuals as compared to allele del.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
