About Netilmicin

What it is

Netilmicin is a semi-synthetic aminoglycoside antibiotic related to gentamicin. It binds bacterial ribosomes and makes them misread genetic instructions, and is used by injection for serious Gram-negative infections and as eye drops, in Europe and other regions; it is not marketed in the US.

How genes come into it

As for all aminoglycosides, the key gene is MT-RNR1, in the DNA of mitochondria, inherited from the mother. Variants such as m.1555A>G make aminoglycosides bind the mitochondrial ribosome and can cause permanent hearing loss. CPIC has published a guideline on MT-RNR1 and aminoglycosides, which covers netilmicin. Most consumer DNA chips do not read these mitochondrial positions reliably.

Published guidelines

Clinical guidelines that name a gene for Netilmicin. They sort people into groups by the gene copies they carry.

CPIC guideline: MT-RNR1. normal risk of aminoglycoside-induced hearing loss · increased risk of aminoglycoside-induced hearing loss · uncertain risk of aminoglycoside-induced hearing loss. Strong. 2022.
