Flupenthixol (Fluanxol)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Flupenthixol

What it is

Flupentixol (flupenthixol) is a first-generation thioxanthene antipsychotic that blocks dopamine D1 and D2 receptors. At higher doses and as a long-acting injection it is used for schizophrenia; at low doses, in some countries, for depression, sometimes combined with melitracen. It is sold as Fluanxol and Depixol, and is not marketed in the US.

How genes come into it

Flupentixol is broken down in the liver, partly by CYP2D6. The gene studies listed, from China and mostly concerning the flupentixol-melitracen combination, looked at NFKB1. The findings come from a few studies, and there is no CPIC or DPWG guideline.

Response (3 findings)

Studies of how well it worked in people carrying a variant.

NFKB1 rs230493: Genotype AT is associated with increased response to flupenthixol in people with Schizophrenia as compared to genotype TT.

NFKB1 rs230504: Genotypes CT + TT is associated with increased response to flupenthixol in people with Schizophrenia as compared to genotype CC.

NFKB1 rs3774959: Genotype AG is associated with increased response to flupenthixol in people with Schizophrenia as compared to genotype GG.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
