Ergot alkaloids are a class of medicines derived from substances made by the ergot fungus. They act on serotonin, dopamine and adrenaline receptors. Members include ergotamine and dihydroergotamine for migraine attacks, methylergometrine to stop bleeding after childbirth, and bromocriptine and cabergoline, which lower prolactin and treat Parkinson's disease. This page collects studies on the class as a group.
The few gene studies listing the class looked at genes of pain and reward signalling (OPRM1, COMT, DRD2, HTR2A, BDNF, DBH), mostly in the context of medication-overuse headache. The findings are weak. Most ergot alkaloids are broken down by CYP3A4, which is why strong CYP3A4 blockers can raise them to dangerous levels; that interaction concerns other medicines, not genes. There is no CPIC or DPWG guideline.

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.
OPRM1 rs1799971: Level 3. Other: Substance Withdrawal Syndrome. 1.
DBH rs1611115: Level 3. Other: Substance Withdrawal Syndrome. 1.
BDNF rs6265: Level 3. Other. 1.
DRD2 rs6275: Level 3. Other. 1.
