Empagliflozin (Jardiance)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Empagliflozin

What it is

Empagliflozin is an SGLT2 inhibitor: it blocks the transporter in the kidney that takes glucose back from the urine, so excess sugar leaves the body. It is used for type 2 diabetes, and, regardless of diabetes, for heart failure and chronic kidney disease, where it reduces hospital admissions and slows decline. It is sold as Jardiance, and with linagliptin or metformin in combinations.

How genes come into it

Empagliflozin is cleared mainly by UGT enzymes and the kidneys. The gene studies on it looked at SLC5A2, the gene for the SGLT2 transporter it blocks, for how much glucose the kidneys passed and how much blood sugar fell. The findings come from a few studies, and there is no CPIC or DPWG guideline.

Response (7 findings)

Studies of how well it worked in people carrying a variant.

ATP2A2 rs1860561: Genotype GG is associated with increased clinical benefit to empagliflozin in people with Heart Failure as compared to genotypes AA + AG.

BDNF rs6265: Genotype CC is associated with increased clinical benefit to empagliflozin in people with Heart Failure as compared to genotypes CT + TT.

SLC5A2 rs11646054: Allele C is not associated with response to empagliflozin in people with Diabetes Mellitus, Type 2 as compared to allele G.

SLC5A2 rs3116149: Allele A is not associated with response to empagliflozin in people with Diabetes Mellitus, Type 2 as compared to allele G.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
