Dobutamine (Dobutrex)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Dobutamine

What it is

Dobutamine is a synthetic catecholamine that mainly activates beta-1 receptors in the heart, making it pump more strongly. Given by continuous drip in hospital, it supports the heart in cardiogenic shock and severe heart failure, and is used in stress echocardiography to test how the heart copes with extra work. It was sold as Dobutrex.

How genes come into it

Dobutamine is broken down within minutes by COMT and conjugation. The gene studies on it looked at ADRB1, the beta-1 receptor gene (its Arg389Gly variant), and GNAS, which carries the receptor's signal, for how strongly the heart rate and pumping responded. The findings come from a few studies, and there is no CPIC or DPWG guideline.

Rated evidence (2 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

GNAS rs62205366: Level 3. Efficacy. 1.

ADRB1 rs1801253: Level 3. Efficacy. 1.

Response (6 findings)

Studies of how well it worked in people carrying a variant.

ADRB1 rs1801253: Genotype CC is associated with increased response to dobutamine in healthy individuals as compared to genotypes CG + GG.

ADRB1 rs1801253: Genotype CC is associated with increased response to dobutamine in healthy individuals as compared to genotypes CG + GG.

ADRB1 rs1801253: Genotype CC is associated with increased response to dobutamine in healthy individuals as compared to genotype GG.

GNAS rs62205366: Genotypes CC + CT are associated with increased response to dobutamine in healthy individuals as compared to genotype TT.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
