Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.
Bortezomib was the first proteasome inhibitor: it blocks the proteasome, the cell's machine for breaking down unwanted proteins, so faulty proteins pile up and myeloma cells die. Given by injection under the skin, it is a backbone of treatment for multiple myeloma and is used for mantle cell lymphoma. Nerve damage is its main side effect. It is sold as Velcade.
Bortezomib is broken down by several CYP enzymes, mainly CYP3A4 and CYP2C19. The gene study listed looked at CTLA4 for side effects. Genome-wide studies have also searched for genes linked to its nerve damage, without a confirmed result. There is no CPIC or DPWG guideline.

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.
CTLA4 rs4553808: Level 3. Toxicity. 1.
Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
