Axitinib is a targeted cancer medicine, a tyrosine kinase inhibitor that selectively blocks the VEGF receptors tumours use to grow new blood vessels. It is used for advanced kidney cancer, alone after earlier treatment or together with an immunotherapy such as pembrolizumab. It is sold as Inlyta.
Axitinib is broken down mainly by CYP3A4 and CYP3A5, with smaller roles for CYP1A2, CYP2C19 and UGT1A1, and carried by ABCB1 and ABCG2. Studies have looked at variants in these genes for its blood levels, and at HIF1A for response. The findings come from a few studies, and there is no CPIC or DPWG guideline.
