Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.
Amantadine was first an antiviral against influenza A and was then found to help Parkinson's disease, where it releases dopamine and blocks NMDA glutamate receptors. Today it is used mainly for Parkinson's disease, especially to reduce involuntary movements (dyskinesia) caused by levodopa, and for some drug-induced movement problems; flu viruses are now largely resistant to it. It is sold as Symmetrel and Gocovri, among other names.
Amantadine is cleared unchanged by the kidneys. The single gene study listed, in Parkinson's disease, looked at SLC22A1 (OCT1), an organic cation transporter, for dose needs and side effects. The evidence is weak, and there is no CPIC or DPWG guideline.

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.
