Agomelatine is an antidepressant that activates melatonin MT1 and MT2 receptors and blocks serotonin 5-HT2C receptors, which is thought to help reset the body clock and raise dopamine and noradrenaline in the frontal brain. It is used for major depression in Europe and Australia; it is not approved in the US. Liver tests are checked because it can raise liver enzymes. It is sold as Valdoxan.
Agomelatine is broken down almost entirely by CYP1A2, so its levels vary widely and are much higher with CYP1A2 blockers such as fluvoxamine. The gene studies listed looked at ABCB1 for response and levels. The findings come from a few studies, and there is no CPIC or DPWG guideline.
