Abatacept (Orencia)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Abatacept

What it is

Abatacept is a biologic medicine built from the CTLA-4 protein fused to part of an antibody. It blocks the second signal T cells need to become fully active (co-stimulation), calming the immune attack. Given by drip or injection, it is used for rheumatoid arthritis, juvenile idiopathic arthritis and psoriatic arthritis, and to prevent graft-versus-host disease. It is sold as Orencia.

How genes come into it

As a protein, abatacept is not broken down by drug-processing enzymes. Studies have looked at the HLA-DRB1 'shared epitope', the main inherited risk factor for rheumatoid arthritis, linked in some studies to a better response, and at the antibody receptor FCGR2A. The findings come from a few studies, and there is no CPIC or DPWG guideline.

HLA-DRB1 HLA-DRB1*04:01, HLA-DRB1*04:04, HLA-DRB1*04:05, HLA-DRB1*04:10, HLA-DRB1*10:01: HLA-DRB1 *04:01 + *04:04 + *04:05 + *04:10 + *10:01 is associated with increased clinical benefit to abatacept in people with Arthritis, Rheumatoid.

HLA-DRB1 HLA-DRB1*04:05: HLA-DRB1 *04:05 is associated with increased clinical benefit to abatacept in people with Arthritis, Rheumatoid.
