Venetoclax (Venclexta)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Venetoclax

What it is

Venetoclax is a BCL-2 inhibitor: it blocks BCL-2, a protein that keeps many blood cancer cells from dying, so they self-destruct. It is used for chronic lymphocytic leukaemia and, with azacitidine or low-dose cytarabine, for acute myeloid leukaemia in people who cannot have intensive chemotherapy. Treatment starts slowly to avoid tumour lysis syndrome. It is sold as Venclexta and Venclyxto.

How genes come into it

Venetoclax is broken down mainly by CYP3A4 (with CYP3A5), so strong CYP3A4 blockers raise its levels many-fold. The gene study listed looked at CYP3A5 for its levels. Resistance arises from acquired changes in BCL2 and related genes in the cancer cells. There is no CPIC or DPWG guideline.
