Tezacaftor

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Tezacaftor

What it is

Tezacaftor is a CFTR corrector: it helps faulty CFTR protein fold properly and reach the cell surface. It is given combined with ivacaftor (Symdeko, Symkevi) and, together with elexacaftor and ivacaftor, in the triple combination Trikafta (Kaftrio), for cystic fibrosis.

How genes come into it

Tezacaftor works only for cystic fibrosis caused by particular variants in CFTR, the gene behind the condition, such as F508del and certain residual-function variants listed in the US label. All studies on this page concern CFTR variants. It is broken down mainly by CYP3A4. There is no CPIC or DPWG guideline for tezacaftor itself.

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

CFTR rs75527207: Level 3. Efficacy: Cystic Fibrosis. 1.

Response (4 findings)

Studies of how well it worked in people carrying a variant.

CFTR rs113993960: Genotype del/del is associated with response to ivacaftor and tezacaftor in people with Cystic Fibrosis.

CFTR rs113993960: Genotype del/del is associated with increased response to ivacaftor and tezacaftor in people with Cystic Fibrosis.

CFTR rs113993960: Genotype CTT/del is associated with increased response to ivacaftor and tezacaftor in people with Cystic Fibrosis.

CFTR rs75527207: Allele A is associated with response to ivacaftor and tezacaftor in people with Cystic Fibrosis.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
