Mitoxantrone (Novantrone)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Mitoxantrone

What it is

Mitoxantrone is an anthracenedione, a chemotherapy medicine related to the anthracyclines: it slips between DNA strands and blocks topoisomerase II. It is used for acute myeloid leukaemia and advanced prostate cancer, and, because it also suppresses immune cells, for aggressive forms of multiple sclerosis. Its use is limited by heart damage over a lifetime total. It is sold as Novantrone, among other names.

How genes come into it

Mitoxantrone is broken down in the liver and pumped out of cells by ABCG2 and ABCB1. Studies have looked at GALNT14 and SLCO1B1 for response in liver cancer, and at a few genes (such as MECP2) for response in multiple sclerosis. The findings come from single or a few studies, and there is no CPIC or DPWG guideline.

Rated evidence (5 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

GALNT14 rs9679162: Level 3. Efficacy: Hepatocellular Carcinoma, Liver cancer. 2.

SLCO1B1 rs2291075: Level 3. Efficacy: Leukemia, Myeloid, Acute. 1.

GALNT14 rs12613732: Level 3. Efficacy: Hepatocellular Carcinoma. 1.

Response (19 findings)

Studies of how well it worked in people carrying a variant.

rs4932551: Allele C is associated with response to cisplatin, fluorouracil and mitoxantrone in people with Carcinoma, Hepatocellular as compared to allele G.

rs5906072: Allele T is associated with response to cisplatin, fluorouracil and mitoxantrone in people with Carcinoma, Hepatocellular as compared to allele C.

rs5934731: Allele C is associated with response to cisplatin, fluorouracil and mitoxantrone in people with Carcinoma, Hepatocellular as compared to allele T.

rs6025211: Genotypes CC + TT are associated with increased response to cisplatin, fluorouracil and mitoxantrone in people with Liver Neoplasms as compared to genotype CT.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
