Mitotane (Lysodren)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Mitotane

What it is

Mitotane is an adrenolytic medicine, related to the insecticide DDT, that destroys cells of the adrenal cortex and blocks steroid hormone production. It is used for adrenocortical carcinoma, a rare adrenal cancer, after surgery or when it has spread, and sometimes for Cushing's syndrome. Blood levels are monitored, and steroid replacement is needed. It is sold as Lysodren.

How genes come into it

Mitotane builds up in fat and strongly speeds up CYP3A4, lowering the levels of many other medicines. The gene study listed linked a CYP2B6 variant (*6) to reaching higher mitotane levels. The evidence is weak, and there is no CPIC or DPWG guideline.
