Flucytosine (5-FC) is an antifungal: fungal cells turn it into fluorouracil, which disrupts their DNA and RNA. It is used together with amphotericin B for serious fungal infections, above all cryptococcal meningitis, and for some severe Candida infections. It is sold as Ancobon, among other names.
Some of the fluorouracil that flucytosine forms reaches the human body, where it is broken down by the enzyme made by DPYD. People with low-function DPYD variants clear fluorouracil poorly, and serious side effects have been reported in them. DPWG has published a guideline on DPYD and flucytosine. Few studies have looked at genes for this medicine.
Clinical guidelines that name a gene for Flucytosine. They sort people into groups by the gene copies they carry.
DPWG guideline: DPYD. DPYD activity score 0 to 2.
