Etanercept (Enbrel)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Etanercept

What it is

Etanercept is a biologic medicine built from the TNF receptor fused to part of an antibody; it acts as a decoy that soaks up TNF, a key signal of inflammation. Injected under the skin once or twice a week, it is used for rheumatoid arthritis, juvenile arthritis, psoriatic arthritis, ankylosing spondylitis and psoriasis. It is sold as Enbrel and as biosimilars.

How genes come into it

Etanercept is not broken down by drug-processing enzymes. The most studied genetic finding is the TNF promoter variant rs1800629 (-308G>A), linked in pooled analyses to a weaker response to TNF blockers in arthritis and psoriasis. Studies have also looked at the TNF receptor genes, CD84 and the antibody receptor FCGR3A. The findings are modest, and there is no CPIC or DPWG guideline.

Response (95 findings)

Studies of how well it worked in people carrying a variant.

rs12081765: Allele G is associated with increased response to adalimumab, certolizumab pegol, etanercept, infliximab or Tumor necrosis factor alpha (TNF-alpha) inhibitors in people with Arthritis, Rheumatoid as compared to allele A.

rs1350948: Allele A is associated with increased response to adalimumab, certolizumab pegol, etanercept, infliximab or Tumor necrosis factor alpha (TNF-alpha) inhibitors in people with Arthritis, Rheumatoid as compared to allele G.

ALPL rs885814: Allele T is associated with decreased response to adalimumab, certolizumab pegol, etanercept, infliximab or Tumor necrosis factor alpha (TNF-alpha) inhibitors in people with Arthritis, Rheumatoid as compared to allele C.

ATP5F1E rs1059150: Genotype GG is associated with decreased response to adalimumab, certolizumab pegol, etanercept, glucocorticoids, infliximab or methotrexate in women with Arthritis, Rheumatoid as compared to genotype TT.
