Daptomycin (Cubicin)

Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Daptomycin

What it is

Daptomycin is a lipopeptide antibiotic that inserts into the membrane of Gram-positive bacteria and disrupts it. Given once daily by drip, it treats complicated skin infections and bloodstream infections, including heart valve infections, caused by Staphylococcus aureus, including MRSA. Muscle damage is a known side effect, so muscle enzymes are checked. It is sold as Cubicin.

How genes come into it

Daptomycin is not broken down by liver enzymes; the kidneys clear it largely unchanged. The gene studies listed looked at ABCB1 variants for its blood levels. The findings come from a few studies, and there is no CPIC or DPWG guideline.

Rated evidence (1 rated variant)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

ABCB1 rs1045642: Level 3. Metabolism/PK. 1.

Metabolism (2 findings)

Studies of how quickly the body processes this medication.

ABCB1 rs1045642: Genotype AA is associated with increased concentrations of daptomycin as compared to genotypes AG + GG.

ABCB1 rs1045642: Genotype AA is associated with decreased clearance of daptomycin as compared to genotypes AG + GG.

Study findings and evidence levels: PharmGKB (CC BY-SA 4.0). Guidelines: CPIC and DPWG, as bundled in PharmCAT. Most studies were done mainly in people of European ancestry.
