Raw research findings for this medication. Mostly single studies, not curated or confirmed robust.

About Clavulanate

What it is

Clavulanic acid (clavulanate) is a beta-lactamase inhibitor: it has little antibacterial effect on its own, but it disables the beta-lactamase enzymes some bacteria use to destroy penicillins. It is combined with amoxicillin (co-amoxiclav, Augmentin) so the antibiotic works against more bacteria, for ear, sinus, chest, skin, dental and bite infections.

How genes come into it

Amoxicillin-clavulanate is one of the most common causes of drug-induced liver injury, usually a few weeks after starting, and the clavulanate part is thought to be mainly responsible. Studies have linked immune-system gene versions, such as HLA-DRB1*15:01 and HLA-DQB1*06:02, and HLA-A*02:01 and HLA-B, to a higher risk. The findings come from a few studies, and there is no CPIC or DPWG guideline.

Rated evidence (2 rated variants)

PharmGKB weighs all studies of a variant and rates how strong the evidence is, from 1A (strongest) to 4.

HLA-DQB1 rs9274407: Level 3. Toxicity: Drug Toxicity. 1.

HLA-B HLA-B*18:01: Level 3. Toxicity. 1.
